Function

- Cytokines that activate STAT3 include growth hormone, IL-6 family cytokines, and G-CSF. STAT3, as well as STAT5, induces progression through the cell cycle, prevents apoptosis and may be associated with cancer development in some cases.

- STAT3 plays an important role in normal development, particularly hematopoiesis. The importance of STAT3 is underscored by the failure of mice lacking STAT3 to survive embryogenesis.

- Crosstalk from pathways other than JAK kinases (JAKs) also leads to phosphorylation and activation of STAT3 as indicated by a role of mTOR (mammalian target of rapamycin, or p70 S6 kinase) and mitogen-activated protein kinase (MAP kinases or MAPKs) pathways in STAT3 activation and signaling.

Pathology

- germline mutations in STAT3-associated hyper-IgE syndrome (STAT3-associated HIES)

  • hyper-IgE syndrome (MIM.147060) or autosomal dominant hyper-IgE recurrent infection syndrome / Job-Buckley syndrome

- Constitutive activation of STAT3 (signal transducer and activator of transcription) has been reported in several primary cancers and tumor cell lines where it induces cell transformation through a combined inhibition of apoptosis and cell-cycle activation.

- STAT3 prevents cell-cycle arrest and cell death through upregulation of survival proteins and downregulation of tumor suppressors.

- STAT-3 expression correlates with lymph node metastasis and cell survival in gastric cancer. (#21072904#)
